XeliGen Lyo PCR Setup

The XeliGen Lyo PCR Setup MyraScript automates preparation of XeliGen lyophilised PCR strip tubes used for HLA genetic testing associated with coeliac disease susceptibility. The workflow reduces manual liquid handling by automating sample and NTC setup, improving reproducibility, consistency, and laboratory efficiency while minimising pipetting errors and consumable usage. The protocol supports up to 18 samples on a Myra and up to 24 samples on a Myra9 instrument.

Two workflow options are available to support different laboratory preferences. The ‘Intermediate Mix’ workflow prepares individual intermediate mixes containing both Resuspension Buffer and Sample/NTC prior to loading into the XeliGen Lyo Strip Tubes, while the ‘Direct-To-Lyo’ workflow transfers Sample/NTC directly from source tubes into the XeliGen Lyo Strip Tubes. Both workflows provide a flexible and scalable approach to PCR setup while maintaining compatibility with the XeliGen Lyo kit.
