PCR Setup with Indexes

This protocol describes the automated setup of PCR reactions using a Myra Liquid Handling System for up to 96 samples. The workflow is designed to support PCR based workflows where unique Index Adapters are incorporated into samples, such as Next Generation Sequencing (NGS) Library Preparation. The workflow distributes either a PCR Master Mix prepared on the instrument or a Premade PCR Master Mix into a PCR reaction plate, adds a unique Index Adapter to each reaction, and optionally transfers samples from a source plate. Samples may also be preloaded into the PCR reaction plate before the
run.

The script uses the sample name and Index Adapter Well ID entered in the Samples Table to associate each sample with its corresponding index. Each unique Index Adapter combination can therefore only be assigned to one sample within a run, ensuring that each sample receives a unique
index combination.
