MIC Broth Microdilution

Broth microdilution is the reference method for assessing the antimicrobial susceptibility of
microorganisms. A series of decreasing antimicrobial concentrations is prepared in growth medium,
followed by the addition of microbial inoculum. After incubation, the MIC is determined as the
lowest antimicrobial concentration that completely inhibits visible microbial growth.

The MIC Broth Microdilution script automates the preparation of individual serial dilution series for
each antimicrobial compound in either a 96 well or 384 well assay plate, with optional inoculum
dispensing to complete assay setup. Multiple compounds, inoculum types and replicate dilution
series can be prepared within a single run, providing flexibility for a wide range of experimental
designs.

Serial dilution series, transfer volumes and reagent requirements are calculated automatically from
the concentrations and dilution parameters entered by the user, eliminating manual calculations and
improving consistency between experiments. The script supports preparation of the starting
concentration directly from the stock solution or, when appropriate, the use of the stock solution as
the first point in the dilution series.

By automating the pipetting intensive steps of assay preparation, the Myra reduces hands on time,
minimises manual pipetting errors and delivers a consistent, accurate and reproducible assay setup
while remaining fully compatible with standard broth microdilution workflows.
