DNA bisulfite conversion for Self-screen methylation assays

This protocol is used for a semi-automated bisulfite conversion workflow, integrating sample concentration, DNA bisulfite conversion, sample clean up and PCR setup into a single workflow for downstream testing with Self screen methylation specific PCR assays, including PreCursor-M+, PreCursor-M AnoGYN and PreCursor-M Gold.

The workflow can process up to 24 samples per run and is intended for genomic DNA using reagents from the EZ 96 DNA Methylation Lightning MagPrep Kit, together with additional MagBinding Buffer, supplied by Zymo Research, Irvine, USA.

The Myra protocol improves workflow consistency, reduces hands on-time and minimises operatordependent variability, requiring only a standard external 96-well thermal cycler for the bisulfite conversion step. Software-guided prompts provide clear, step by step instructions throughout the workflow, from deck setup through to selection and initiation of the appropriate thermal cycler program, supporting a consistent and reliable workflow from sample input through to PCR setup.
