The Ligation Sequencing Kit V14 is used for the preparation of DNA libraries for next-generation sequencing (NGS) on Oxford Nanopore Technologies platforms. It streamlines the process of preparing DNA samples for sequencing, making it more efficient and accurate. This kit enhances the yield of DNA from limited starting material, reduces biases that can skew sequencing results, and ensures compatibility with various types of DNA samples. This protocol semi-automates Ligation Sequencing Kit V14 for up to 24 libraries on the Myra where the 96 Well Magnetic Station is used for the clean-up steps and the heating steps are performed on an external thermal cycler. By automating the pipetting-intensive steps, human error can be eliminated, thereby providing high-quality reproducible results, and increasing overall productivity.
